Therapeutics of Natural and Synthetic Compounds in Protease-Induced Cancer
Abstract
Colorectal cancer (CRC) is a leading cause of cancer mortality worldwide. There is an immediate need for therapeutic approaches. This chapter focuses on the proteasome, a key enzyme complex crucial for protein degradation, and its role in CRC. It describes how the ubiquitin-proteasome system modulates key signaling pathways, such as Wnt/β-catenin, to regulate processes necessary to the development of CRC, such as cell proliferation and apoptosis. Proteasome inhibitors (PIs) are presented as promising therapeutic agents capable of inducing apoptosis and inhibiting tumor growth by interfering with these processes. The text navigates through the potential of PIs in CRC therapy, especially in combination treatments, acknowledging the challenges faced, such as drug resistance and adverse effects, particularly in combination regimens. In the end, it seeks to improve patient outcomes in CRC management by highlighting the necessity of ongoing research and development to increase the effectiveness of PI-based therapies. © 2025 Elsevier Inc.
